Abstract
The present invention relates to a cell, wherein the cell is modified to: reduce O-GalNAc galactosylation activity in the cell by reduction of functional COSMC molecular chaperone in the cell and/or by reduction of functional T-synthase in the cell; and overexpress β1,4-galactosyltransferase in the cell; and associated methods, kits and uses.
Claims22
A cell, wherein the cell is modified to:
- reduce O-GalNAc galactosylation activity in the cell by reduction of functional COSMC molecular chaperone in the cell and/or by reduction of functional T-synthase in the cell;
- and
- overexpress β1,4-galactosyltransferase in the cell.
(canceled)
A method of modifying a cell, wherein the cell is modified to enhance β-1,4 galactosylation of a polypeptide product, the modifications comprising:
- reducing O-GalNAc galactosylation activity in the cell by reduction of functional COSMC molecular chaperone in the cell and/or by reduction of functional T-synthase in the cell;
- and
- modification of the cell to provide overexpression of β1,4-galactosyltransferase in the cell.
18-20. (canceled)
A plasmid encoding:
- a genetic modification element(s) that is targeted to knock-out or reduce functional COSMC molecular chaperone and/or T-synthase expression in a cell;
- b4GalT for expression in the cell;
- and
- optionally a selection marker, such as an antibiotic resistance gene.
A kit comprising:
- a first plasmid encoding:
- a genetic modification element(s) that is targeted to knock-out or reduce functional COSMC molecular chaperone and/or T-synthase expression in a cell;
- and
- a second plasmid encoding b4GalT for expression in a cell and optionally a selection marker, such as an antibiotic resistance gene.
