Abstract
The disclosure provides inducible caspase polypeptides, compositions comprising inducible caspase polypeptides and sequences encoding the same, cells modified to express the polypeptides and compositions of the disclosure, as well as methods of making and methods of using same for adoptive cell therapy.
Claims9
A method of modifying a cell therapy in a subject in need thereof, comprising administering to the subject (a) an inducible caspase polypeptide comprising the amino acid sequence of SEQ ID NO:
- 9 or (b) a polynucleotide comprising the nucleic acid sequence of SEQ ID NO:
- 10,
- wherein apoptosis may be selectively induced in the cell by contacting the cell with an induction agent.
A method of treating a disease or disorder in a subject in need thereof comprising
- (a) administering to the subject a cell comprising (i) a kinetic agent and (ii) an inducible caspase polypeptide comprising the amino acid sequence of SEQ ID NO:
- 9 or a polynucleotide comprising the nucleic acid sequence of SEQ ID NO:
- 10,
- wherein apoptosis may be selectively induced in the cell by contacting the cell with an induction agent, and wherein the cell comprising the kinetic agent induces local tissue toxicity within a target tissue of the subject, and
- (b) selectively inducing apoptosis in the cell comprising the kinetic agent prior to induction of significant toxicity in a non-target tissue of the subject, thereby eliminating the target tissue, preserving non-target tissue and treating the disease or disorder in the subject.
