Abstract
A system for buoyant-particle-assisted cell therapy includes and/or interfaces with a set of buoyant particles. Additionally or alternatively, the system can include and/or interface with a processing container, a set of processing materials (e.g., buffers, factors, solutions, etc.), and/or any other components. A method for buoyant-particle-assisted cell therapy includes processing the set of cells of interest. Additionally or alternatively, the method can include any or all of: preparing a set of buoyant particles; receiving a sample; and isolating a set of cells of interest from the sample.
Claims21
A method for buoyancy-assisted cell therapy manufacturing, the method comprising:
- initiating a 1st selection process at a 1st container, the 1st selection process comprising:
- receiving a 1st sample comprising a set of target materials at the 1st container, the set of target materials comprising a set of T cells, wherein a remainder of the 1st sample comprises a set of non-target materials;
- introducing a 1st set of buoyant particles to the 1st sample, the 1st set of buoyant particles configured to bind with the set of non-target materials to produce a set of bound non-target materials, thereby enabling an aggregation of the set of bound non-target materials at a superior region of the 1st sample;
- collecting the set of target materials from an inferior region of the 1st sample;
- initiating a cell activation and expansion process comprising a 2nd selection process at a 2nd container, the cell activation process comprising:
- receiving a 2nd sample comprising the set of target materials at the 2nd container;
- introducing a set of activation factors to the 2nd sample, wherein the set of activation factors comprises multiple types of antibodies, the set of antibodies configured to bind with the set of target materials;
- introducing a 2nd set of buoyant particles to the 2nd sample, the 2nd set of buoyant particles configured to bind with the set of activation factors to produce a set of bound target materials, wherein in at least a portion of the set of bound target materials, a single T cell is bound to multiple buoyant particles of the 2nd set of buoyant particles;
- processing the set of bound target materials, comprising:
- immobilizing the set of T cells to surfaces of the 2nd set of buoyant particles through the set of activation factors, thereby facilitating activation of the set of T cells;
- facilitating aggregation of the set of bound target materials at a superior region of the 2nd sample;
- facilitating expansion of the set of T cells to form an expanded set of T cells;
- and
- facilitating a spatial separation of the expanded set of T cells relative to the 2nd set of buoyant particles and the set of activation factors, thereby preventing exhaustion of the expanded set of T cells.

- facilitating aggregation of the set of bound target materials at a superior region of a container cavity, wherein the set of bound target materials is arranged in the container cavity;
- facilitating expansion of the set of T cells to form an expanded set of T cells;
- and
- facilitating a spatial separation of the expanded set of T cells relative to the 2nd set of buoyant particles and the set of activation factors, thereby preventing exhaustion of the expanded set of T cells.
