Abstract
Provided herein are methods for treating at least one symptom of ALS in a subject who has received a first dosage of a substrate of a CYP or a transporter, the method including administering to the subject a composition comprising a bile acid or a pharmaceutically acceptable salt thereof and a phenylbutyrate compound, monitoring the subject for response to the substrate, and administering a second dosage of the substrate, wherein the second dosage is less than the first dosage.

A method of treating at least one symptom of Amyotrophic Lateral Sclerosis (ALS) in a subject, the method comprising:
- (a) administering to a subject who has received a first dosage of a substrate of a cytochrome P450 (CYP) a composition comprising about 1 gram of Taurursodiol (TURSO) and about 3 grams of sodium phenylbutyrate,
- (b) determining or having determined a first level of the substrate or a metabolite thereof in a first biological sample from the subject, and
- (c) administering to the subject a second dosage of the substrate, wherein the second dosage is lower than the first dosage.

A method of treating at least one symptom of ALS in a subject who has received a first dosage of a substrate of Organic Anion Transporter 1 (OAT1), the method comprising:
- (a) determining or having determined a first level of the substrate or a metabolite thereof in a first biological sample from the subject;
- (b) administering to the subject a composition comprising TURSO and sodium phenylbutyrate;
- (c) determining or having determined a second level of the substrate or the metabolite thereof in a second biological sample from the subject, wherein the second level is higher than the first level;
- and
- (d) administering to the subject a second dosage of the substrate, wherein the second dosage is lower than the first dosage.
A method of administering a composition comprising TURSO and sodium phenylbutyrate to a subject who has received a first dosage of a substrate of OAT1, the method comprising:
- (a) determining or having determined a first level of the substrate or a metabolite thereof in a first biological sample from the subject;
- (b) administering to the subject a composition comprising TURSO and sodium phenylbutyrate;
- (c) determining or having determined a second level of the substrate or the metabolite thereof in a second biological sample from the subject, wherein the second level is higher than the first level;
- and
- (d) administering to the subject a second dosage of the substrate, wherein the second dosage is lower than the first dosage.
