Abstract
The present invention aims to increase cytotoxic activity of an immunocompetent cell to thereby enhance a therapeutic effect against diseases such as cancers, and provides an immunocompetent cell having decreased diacylglycerol kinase activity, which cell expresses a fusion protein including IL-15 and an IL-15 receptor α subunit, and which cell expresses a chimeric antigen receptor.
Claims21
An immunocompetent cell having decreased diacylglycerol kinase activity, wherein said cell expresses a fusion protein (IL-15/IL-15Rα) comprising IL-15 and an IL-15 receptor α subunit (IL-15Rα), and expresses a chimeric antigen receptor.

(canceled)
A method of producing an immunocompetent cell that expresses a chimeric antigen receptor, comprising the step of:
- (a) providing an immunocompetent cell that expresses a chimeric antigen receptor;
- wherein the immunocompetent cell has decreased diacylglycerol kinase activity, and expresses a fusion protein (IL-15/IL-15Rα) comprising IL-15 and an IL-15 receptor α subunit (IL-15Rα).
