Abstract
The present disclosure relates to a variant polypeptide having attenuated dihydrodipicolinate reductase activity and a method of producing L-threonine using the same.
Claims15
A variant polypeptide having dihydrodipicolinate reductase activity, wherein an amino acid at position 13 in an amino acid sequence of SEQ ID NO:
- 1 or an amino acid sequence having 98% or more identity thereto is substituted with asparagine, threonine, cysteine, tyrosine, serine, lysine, or glutamine, and the variant polypeptide has at least 80% or more and less than 100% sequence homology to the amino acid sequence of SEQ ID NO:
- 1 or the amino acid sequence having 98% or more identity thereto.
A microorganism of the genus Corynebacterium (Corynebacterium sp.) comprising any one or more of a variant polypeptide having dihydrodipicolinate reductase activity, wherein an amino acid at position 13 in an amino acid sequence of SEQ ID NO:
- 1 or an amino acid sequence having 98% or more identity thereto is substituted with asparagine, threonine, cysteine, tyrosine, serine, lysine, or glutamine, and the variant polypeptide has at least 80% or more and less than 100% sequence homology to the amino acid sequence of SEQ ID NO:
- 1 or the amino acid sequence having 98% or more identity thereto;
- a polynucleotide encoding the polypeptide;
- and a vector including the polynucleotide.
A method of producing L-threonine, the method comprising the step of culturing, in a medium, a microorganism of the genus Corynebacterium (Corynebacterium sp.) including a variant polypeptide having dihydrodipicolinate reductase activity, wherein an amino acid at position 13 in an amino acid sequence of SEQ ID NO:
- 1 or an amino acid sequence having 98% or more identity thereto is substituted with asparagine, threonine, cysteine, tyrosine, serine, lysine, or glutamine, and the variant polypeptide has at least 80% or more and less than 100% sequence homology to the amino acid sequence of SEQ ID NO:
- 1 or the amino acid sequence having 98% or more identity thereto.
