Abstract
Disclosed herein are compositions and uses thereof for detecting HIV latency reversal, isolating cells with HIV latency reversal, treating HIV infection, and/or reversing latency in HIV infected CD4+ T cells. In some aspects, disclosed herein is a composition and uses thereof for treating HIV infection, wherein the composition comprises one or more mature monocyte-derived dendritic cells (MDGs) having an HIV peptide bound to a Class I major histocompatibility complex (MHC) molecule and a herpesvirus peptide bound to one or more Class II MHC molecules.
Claims41
A method of treating a human immunodeficiency virus-1 (HIV-1) infection in a subject, comprising:
- a) obtaining one or more mature monocyte-derived dendritic cells (MDCs) having an HIV-1 peptide bound to a Class I major histocompatibility complex (MHC) molecule and a herpesvirus peptide bound to a Class II MHC molecule;
- and
- b) administering to the subject an effective amount of the one or more MDCs of step a) and thereby treating the HIV-1 infection.

A composition comprising one or more mature monocyte-derived dendritic cells (MDCs) having an HIV-1 peptide bound to a Class I major histocompatibility complex (MHC) molecule and a herpesvirus peptide bound to a Class II MHC molecule.
A method of reversing HIV-1 latency in a CD4+ T cell in a subject, comprising:
- a) obtaining one or more mature monocyte-derived dendritic cells (MDCs) having a herpesvirus peptide bound to a Class II MHC molecule;
- and
- b) administering to the subject an effective amount of the one or more MDCs of step a) and thereby reversing the HIV-1 latency.

A method of detecting HIV-1 latency reversal in a CD4+ T cell comprising:
- a) culturing an HIV-1 latent infected CD4+ T cell with one or more mature monocyte-derived dendritic cells (MDCs) having an HIV-1 peptide and a herpesvirus peptide bound to a Class II MHC molecule;
- and
- b) identifying within the culture a CD4+ T cell having reduced CD4 expression as compared to a control, wherein the identification is the detection of HIV-1 latency reversal in the CD4+ T cell.
