Abstract
Provided are anti-PD-L1 diabodies, imaging agents, methods and kits for determination of the distribution and expression levels of PD-L1 in an individual having a disease or condition. Anti-PD-L1 diabody agents, and methods for treating diseases or disorders are also provided.
Claims48
An isolated anti-PD-L1 diabody comprising a first polypeptide and a second polypeptide, wherein the first polypeptide and the second polypeptide each comprises a heavy chain variable region (VH) and a light chain variable region (VL), wherein:
- a) the VH comprises an HC-CDR1 comprising the amino acid sequence of SEQ ID NO:
- 1, an HC-CDR2 comprising the amino acid sequence of SEQ ID NO:
- 6, and an HC-CDR3 comprising the amino acid sequence of any one of SEQ ID NO:
- 11, or a variant thereof comprising up to a total of about 5 amino acid substitutions in the HC-CDRs;
- and
- b) the VL comprises an LC-CDR1 comprising the amino acid sequence of SEQ ID NO:
- 14, an LC-CDR2 comprising the amino acid sequence of SEQ ID NO:
- 17, and an LC-CDR3 comprising the amino acid sequence of any one of SEQ ID NO:
- 20, or a variant thereof comprising up to a total of about 5 amino acid substitutions in the LC-CDRs.
An isolated anti-PD-L1 diabody comprising a first polypeptide and a second polypeptide, wherein the first polypeptide and the second polypeptide each comprises a heavy chain variable region (VH) and a light chain variable region (VL), and wherein a) the VH and the VL of the first polypeptide, b) the VH and the VL of the second polypeptide, c) the VH of the first polypeptide and the VL of the second polypeptide, or d) the VH of the second polypeptide and the VL of the first polypeptide form a binding domain that specifically binds to PD-L1 competitively with an anti-PD-L1 antibody comprising a second heavy chain variable region (VH-2) and a second light chain variable region (VL), wherein:
- a) the VH-2 comprises an HC-CDR1 comprising the amino acid sequence of SEQ ID NO:
- 1, an HC-CDR2 comprising the amino acid sequence of SEQ ID NO:
- 6, and an HC-CDR3 comprising the amino acid sequence of SEQ ID NO:
- 11;
- and
- b) the VL2 comprises an LC-CDR1 comprising the amino acid sequence of SEQ ID NO:
- 14, an LC-CDR2 comprising the amino acid sequence of SEQ ID NO:
- 17, and an LC-CDR3 comprising the amino acid sequence of SEQ ID NO:
- 20.
An isolated anti-PD-L1 diabody comprising a first polypeptide and a second polypeptide, wherein the first polypeptide and the second polypeptide each comprises a heavy chain variable region (VH) and a light chain variable region (VL), wherein:
- a) the VH comprises a HC-CDR1, a HC-CDR2, and a HC-CDR3, respectively comprising the amino acid sequences of a CDR1, a CDR2, and a CDR3 within a second heavy chain variable region (VH-2) having the sequence set forth in any of SEQ ID NOs:
- 22-24;
- and
- b) the VL comprises a LC-CDR1, a LC-CDR2, and a LC-CDR3, respectively comprising the amino acid sequences of a CDR1, a CDR2, and a CDR3 within a second light chain variable region (VL-2) having the sequence set forth in any of SEQ ID NOs:
- 25-27.
