Abstract
The present invention provides an enhanced osteogenic composition comprising of connective tissue proteins having molecular weights greater than or equal to 3.5 kDa wherein the composition is prepared by treating demineralized bone material in an acidic extraction medium at a pH between about 0.10 to 0.45 at an extraction temperature between greater than 25° C. and less than 80° C. for a predetermined time period. The present invention further provides a method of making the enhanced osteogenic composition.

1-24. (canceled)
A method of preparing an enhanced osteogenic composition comprising of treating demineralized bone material in an acidic extraction medium at a pH between about −0.10 to 0.45 at an extraction temperature between greater than 25° C. and less than 80° C. for a predetermined time period between 48 hours to 120 hours, wherein:
- a) the enhanced osteogenic composition, derived from the demineralized bone material treated in the acidic extraction medium, contains connective tissue proteins with molecular weights greater than or equal to 3.5 kDa;
- and
- b) the connective tissue proteins promote bone growth.
A method of preparing an enhanced osteogenic composition comprising of:
- a) treating demineralized bone material in an acidic extraction medium at a pH between about 0.10 to 0.45 at an extraction temperature between greater than 25° C. and less than 80° C. for a predetermined time period between 48 hours to 120 hours resulting in a viscous liquid composition;
- and
- b) dialyzing the viscous liquid composition with a Regenerated Cellulose Membrane having a pore size greater than or equal to 3.5 kDa resulting in the enhanced osteogenic composition having a pH greater than or equal to 4.5, wherein the enhanced osteogenic composition increases osteogenic activity by providing access to the connective tissue proteins derived from the demineralized bone material treated in the acidic extraction medium.
