Abstract
Genetically modified microorganisms that have the ability to convert carbon substrates into chemical products such as 2,3-BDO are disclosed. For example, genetically modified methanotrophs that are capable of generating 2,3-BDO at high titers from a methane source are disclosed. Methods of making these genetically modified microorganisms and methods of using them are also disclosed.
Claims23
1-31. (canceled)
The vector of claim 147, comprising two or more genes encoding:
- an acetoin reductase, an alpha-acetolactate decarboxylase, and/or an acetolactate synthase.
The vector of claim 147, wherein the gene encoding the acetolactate synthase is 5′ in relation to any other gene on the vector.
The vector of claim 147, wherein the gene encoding the acetoin reductase is 3′ in relation to any other gene on the vector.
The vector of claim 147, wherein the gene encoding the alpha-acetolactate decarboxylase is neither 5′ or 3′ in relation to any other gene on the vector.
The vector of claim 147, comprising a gene encoding an acetoin reductase.
The vector of claim 147, comprising a gene encoding an alpha-acetolactate decarboxylase.
The vector of claim 147, comprising a gene encoding an acetolactate synthase.
The vector of claim 147, further comprising a promoter that is active within a methanotroph.
The vector of claim 147, wherein the vector is an integration vector.
The vector of claim 147, wherein the vector is an episomal vector.
42-139. (canceled)
The vector of claim 147 comprising one or more genes encoding an acetoin reductase, an alpha-acetolactate decarboxylase, and/or an acetolactate synthase, wherein the one or more genes is under the control of a switch.
141-143. (canceled)
A vector comprising:
- (a) two or more genes encoding:
- an acetoin reductase, an alpha-acetolactate decarboxylase, and/or an acetolactate synthase;
- or
- (b) one or more genes encoding an acetoin reductase, an alpha-acetolactate decarboxylase, and/or an acetolactate synthase, wherein the one or more genes is/are under the control of a switch.
