## Abstract

This disclosure relates to antibodies that specifically bind to the Interleukin-13 receptor subunit alpha-2 (IL13Rα2) protein, and associated uses and methods for production.

## Claims

### Claim 1

An antibody that binds to Interleukin-13 receptor subunit alpha-2 (IL13Rα2), which comprises a heavy chain variable region comprising the amino acid sequence of SEQ ID NO: 3.

### Claim 2 (depends on 1)

The antibody according to claim 1, wherein the antibody further comprises a light chain variable region comprising the amino acid sequence of SEQ ID NO: 4.

### Claim 3 (depends on 1)

The antibody according to claim 1, wherein the antibody binds IL13Rα2 with an affinity (KD) no greater than 5×10−10 M.

### Claim 4 (depends on 1)

The antibody according to claim 1, wherein the antibody competitively inhibits the binding to IL13Rα2 of an antibody comprising a heavy chain variable region having the amino acid sequence of SEQ ID NO: 1 and a light chain variable region having the amino acid sequence of SEQ ID NO: 2.

### Claim 5 (depends on 1)

The antibody according to claim 1, wherein the antibody binds IL13Rα2 with higher affinity (KD) than an antibody comprising a heavy chain variable region having the amino acid sequence of SEQ ID NO: 1 and a light chain variable region having the amino acid sequence of SEQ ID NO: 2.

### Claim 6 (depends on 1)

The antibody according to claim 1, wherein the antibody has immunogenicity in a human subject that is lower than that of an antibody comprising a heavy chain variable region having the amino acid sequence of SEQ ID NO: 1 and a light chain variable region having the amino acid sequence of SEQ ID NO: 2.

### Claim 7 (depends on 1)

The antibody according to claim 1, wherein the antibody has improved stability properties as compared to an antibody comprising a heavy chain variable region having the amino acid sequence of SEQ ID NO: 1 and a light chain variable region having the amino acid sequence of SEQ ID NO: 2.

### Claim 8 (depends on 1)

The antibody according to claim 1, wherein the antibody comprises a constant region of isotype IgG1, IgG2, IgG3 or IgG4, or a mutated IgG constant region.

### Claim 9 (depends on 8)

The antibody of claim 8, wherein the constant region is human.

### Claim 10 (depends on 1)

The antibody according to claim 1, wherein the antibody is a Fab, a Fab′, a F(ab′)2, or an scFv.

### Claim 11 (depends on 1)

An antibody drug conjugate comprising the antibody according to claim 1 conjugated to a drug moiety.

### Claim 12 (depends on 11)

The antibody drug conjugate of claim 11, wherein the drug moiety comprises a pyrollobenzodiazepine dimer.

### Claim 13 (depends on 1)

A pharmaceutical composition comprising the antibody of claim 1 and a pharmaceutically acceptable diluent, carrier, or excipient.

### Claim 14 (depends on 1)

A method of treating cancer in a subject, comprising administering the antibody of claim 1 to a subject in need thereof.

### Claim 15 (depends on 11)

A method of treating cancer in a subject, comprising administering the antibody drug conjugate of claim 11 to a subject in need thereof.

### Claim 16 (depends on 13)

A method of treating cancer in a subject, comprising administering the pharmaceutical composition of claim 13 to a subject in need thereof.

### Claim 17 (depends on 1)

A polynucleotide encoding an antibody according to claim 1.

### Claim 18 (depends on 17)

A vector comprising the polynucleotide of claim 17.

### Claim 19 (depends on 18)

A host cell comprising the vector according to claim 18.

### Claim 20 (depends on 11)

A pharmaceutical composition comprising the antibody drug conjugate of claim 11 and a pharmaceutically acceptable diluent, carrier, or excipient.
