Abstract
The disclosure provides systems and methods for droplet processing. For example, a method can include providing a first droplet and a second droplet. In some cases, the first droplet may have a first concentration of a reagent and the second droplet may have a second concentration of the reagent. The second droplet may comprise a bead or a biological particle. The method can also include subjecting the first droplet and the second droplet to conditions sufficient to transfer the reagent from the first droplet to the second droplet, thereby decreasing the first concentration in the first droplet and increasing the second concentration in the second droplet.
Claims32
A method for droplet processing, comprising:
- (a) providing a first droplet population and a second droplet population, wherein droplets from said first droplet population have a first concentration of a reagent and droplets from said second droplet population have a second concentration of said reagent, and wherein said droplets from said second droplet population comprise a bead or an analyte carrier;
- and
- (b) subjecting said first droplet population and said second droplet population to conditions sufficient to transfer said reagent between said first droplet population and said second droplet population, thereby changing said first concentration of said reagent in said first droplet population and said second concentration of said reagent in said second droplet population.

A method for nucleic acid processing, comprising:
- (a) agitating (i) a first fluid volume comprising a population of beads and a population of analyte carriers, wherein said population of beads comprises a plurality of nucleic acid barcode molecules, wherein said population of analyte carriers comprises a plurality of nucleic acid molecules, and (ii) a second fluid volume immiscible with said first fluid volume, to generate a plurality of droplets, wherein a first droplet from said plurality of droplets comprises a bead from said population of beads and an analyte carrier from said population of analyte carriers, wherein said bead comprises a nucleic acid barcode molecule of said plurality of nucleic acid barcode molecules, wherein said analyte carrier comprises a nucleic acid molecule of said plurality of nucleic acid molecules, wherein said first droplet has a first concentration of a reagent, and wherein said reagent is configured to release said nucleic acid barcode molecule or release said nucleic acid molecule in said droplet;
- (b) subjecting said first droplet and a second droplet having a second concentration of said reagent to conditions sufficient to transfer said reagent between said first droplet and said second droplet, thereby changing said first concentration of said reagent in said first droplet and said second concentration of said reagent in said second droplet;
- and
- (c) generating a barcoded nucleic acid molecule using at least said nucleic acid barcode molecule and said nucleic acid molecule in said first droplet.
