Abstract
Provided herein are compositions and methods for analyzing neutralizing antibodies to SARS-CoV-2, for example, in a sample from a subject suspected of having, having, or having had (e.g., having recovered from) a SARS-CoV-2 infection, or a subject having received a prophylactic and/or therapeutic intervention for a coronavirus infection.
Claims26
A method for analyzing a sample, comprising:
- contacting the sample with a particle coated with a surface antigen from a coronavirus, and with a receptor for the surface antigen which is labelled with a detectable moiety,
- wherein the sample comprises or is suspected of comprising an antibody that reduces or inhibits binding between the surface antigen and the receptor,
- wherein labelled receptors that specifically bind to the surface antigen are immobilized on the particle via the surface antigen, wherein the antibody, if present in the sample, competes with or displaces one or more of the labelled receptors, and
- wherein a signal is detected on the particle, and a magnitude of the signal is inversely proportional to an amount or level of the antibody in the sample.

A method for analyzing a sample, comprising:
- contacting the sample with a particle coated with a surface antigen from a coronavirus to provide a reaction mix, wherein the sample comprises or is suspected of comprising an antibody that reduces or inhibits binding between the surface antigen and a receptor for the surface antigen;
- and
- contacting the reaction mix with the receptor which is labelled with a detectable moiety,
- wherein labelled receptors that specifically bind to the surface antigen are immobilized on the particle via the surface antigen, wherein the antibody, if present in the sample, competes with or displaces one or more of the labelled receptors, and
- wherein a signal is detected on the particle, and a magnitude of the signal is inversely proportional to an amount or level of the antibody in the sample.

A kit for analyzing a sample, which kit comprises:
- a) a surface antigen from a coronavirus optionally coated on a bead to form a first assay reagent, optionally wherein the bead is a magnetic bead;
- and
- b) a receptor for the surface antigen conjugated to a chemiluminescent moiety to form a second assay reagent.
