Abstract
Deuterated polyunsaturated fatty acid (“PUFA”) compounds, compositions, and uses of the compounds for reducing lipid autooxidation and the treatment of various diseases and conditions are provided.
Claims50
A method of treating or ameliorating a disease or condition caused by lipid autooxidation in a subject in need thereof, comprising administering an effective amount of a compound of Formula (I), or a pharmaceutically acceptable salt thereof, to the subject:
- or a pharmaceutically acceptable salt thereof, wherein:
- each of Y1, Y2, Y3, Y4, Y5, Y6, Y7, Y8, Y9, Y10, Y11, Y12, Y13, Y14, and Y15 are independently hydrogen and deuterium;
- X1 is —C(Y14Y15)—CH═CH—, —CH2—, —CD2—, or a direct bond;
- X2 is —CH2CH2—, —CH2CD2—, —CD2CH2—, —CD2CD2—, or —CH═CH—;
- R1 is a substituted or unsubstituted —O—C1-C6 alkyl, a substituted or unsubstituted —S—C1-C6 alkyl, a substituted or unsubstituted —NH—C1-C6, alkyl, —NH2, —OH, an unsubstituted sphingolipid, or an unsubstituted glyceryl ester;
- wherein at least two of Y1, Y2, Y3, Y4, Y5, Y6, Y7, Y8, Y9, Y10, Y11, Y12, Y13, Y14, and Y15 are independently deuterium;
- wherein when each of Y1-Y5 are deuterium, then X1 is —C(Y14Y15)—CH═CH— and at least one of Y6—Y15 is deuterium;
- or X1 is —CD2-;
- or X2 is —CH2CD2—, —CD2CH2—, or —CD2CD2-;
- wherein the compound of Formula (I), or a pharmaceutically acceptable salt thereof, comprises from about 1% to about 99% of the total amount of fats, fatty acids, and fatty acid esters administered to, or ingested by, the subject.
A liposomal composition comprising from about 1% to about 99% of one or more deuterated compounds in the liposome bilayer, wherein the one or more deuterated compounds are selected from the group consisting of:
- and salts of any of the foregoing.
A compound of Formula (II), or a pharmaceutically acceptable salt thereof,
- wherein:
- each of Y1A, Y2A, Y3A, Y4A, Y5A, Y6A, Y7A, Y8A, Y9A, Y10A, Y11A, Y12A, Y13A, Y14A, and Y15A are independently hydrogen and deuterium;
- X1A is —C(Y14AY15A)—CH═CH—, —CH2—, —CD2—, or a direct bond;
- X2A is —CH2CH2—, —CD2CD2—, or —CH═CH—;
- R1A is a substituted or unsubstituted —O—C1-C6 alkyl, a substituted or unsubstituted —S—C1-C6 alkyl, a substituted or unsubstituted —NH—C1-C6, alkyl, —NH2, —OH, an unsubstituted sphingolipid, an unsubstituted glyceryl ester, or
- R3A is an unsubstituted C1-C20 alkyl;
- wherein at least two Y1A-Y15A are not hydrogen;
- and
- wherein Formula (II) is not selected from:
- 7,7,10,10,13,13,16,16-D8-eicosapentaenoic acid, or a salt or ester thereof;
- 13,13,16,16-D4-eicosapentaenoic acid, or a salt or ester thereof;
- 19,19,20,20,20-D5-eicosapentaenoic acid, or a salt or ester thereof;
- 21,21,22,22,22-D5-docosahexaenoic acid, or a salt or ester thereof;
- 6,6,9,9,12,12,15,15,18,18-D10-docosahexaenoic acid, or a salt or ester thereof;
- or 7,7,10,10,13,13-D6-arachadonic acid, or a salt or ester thereof.
