Abstract
A method of detecting the presence of a prostate cancer field defect in a human subject. The method includes (a) obtaining genomic DNA from the human subject and (b) determining methylation status in at least one target region selected from the group consisting of PLA2G16, CAV1, EVX1, MCF2L, FGF1, NCR2, WNT2, EXT1, and SPAG4, wherein methylation changes indicate the presence of prostate cancer or a prostate cancer field defect, wherein the change is relative to tissue from a second human subject who does not have prostate cancer.
Claims27
A method of detecting a prostate cancer field defect in a human, the method comprising:
- (a) obtaining a first genomic DNA sample from a first human subject known not to have prostate cancer;
- and
- (b) determining methylation status of the first genomic DNA sample of step (a) in at least one target region selected from the group consisting of PLA2G16, CAV1, EVX1, MCF2L, FGF1, NCR2, WNT2, EXT1, and SPAG4, thereby generating a normal methylation profile;
- (c) repeating steps (a) and (b) with a second genomic DNA sample from a second human subject suspected of having prostate cancer, thereby generating a test methylation profile;
- (d) comparing the test methylation profile to the normal methylation profile, wherein methylation changes in the test methylation profile as compared to the normal methylation profile indicate presence of prostate cancer or a prostate cancer field defect in the second human subject.
A kit for detecting a prostate cancer field defect, the kit comprising:
- at least one pair of polymerase chain reaction primers, wherein the primers are dimensioned and configured to amplify a target region in a sample of human genomic DNA selected from the group consisting of SEQ. ID. NOS:
- 1-6, 18, 39, and 101.
