Abstract
The present disclosure is based on the identification of a novel sequence motif present in halogenase type enzymes. From this, the disclosure provides methods for identifying and or detecting halogenases (halogenating enzymes) and novel halogenases identifiable using such methods. The disclosure also provides a novel cohort of enzymes and novel methods for achieving substrate halogenation.
Claims19
A method of identifying or detecting amino acid sequences which potentially encode or provide halogenases, said method comprising probing or screening an amino acid sequence for the presence of a motif or consensus sequence/motif having the sequence:

- Fx•Px•Sx•G
- or
- (SEQ ID NO:
- 2)
- FxxPxxSxG;
- wherein an amino acid sequence comprising said motif or consensus sequence, is an amino acid sequence which potentially encodes or provides a halogenase.
A method of identifying or detecting nucleic acid sequences which potentially encode halogenases, said method comprising probing or screening an nucleic acid sequence for the presence of a nucleic acid sequence which encodes the motif or consensus sequence/motif having the sequence:

- or
- (SEQ ID NO:
- 2)
- FxxPxxSxG;
- wherein a nucleic acid sequence comprising a sequence which encodes said motif or consensus sequence, is a nucleic acid sequence which potentially encodes a halogenase.
A halogenase encoded or provided by any one of the sequences of SEQ ID NOS:
- 5-149 or a functional fragment, variant or derivative thereof.
A halogenase comprising a sequence of any one of SEQ ID NOS:
- 5-149 or a functional fragment, variant or derivative thereof.
A halogenase comprising the sequence of SEQ ID NO:
- 7 or a functional fragment, variant or derivative thereof, or a nucleic acid sequence encoding SEQ ID NO:

A method of halogenating a substrate, said method comprising contacting a substrate to be halogenated with one or more halogenases comprising any of the sequences provided by SEQ ID NOS:
