Abstract
The present disclosure relates to immunogenic formulations and their use for the treatment and prevention of T. cruzi infection, Chagas Disease, and chronic Chagas cardiomyopathy. In certain aspects, methods of producing antigen presenting cells comprising T. cruzi antigens are provided.
Claims101
An antigen presenting cell comprising the recombinant nucleic acid encoding an expression vector for expression of least a first antigen from an intracellular pathogen.
A recombinant nucleic acid comprising a sequence encoding at least a first antigen from T. cruzi and a sequence encoding a genetic adjuvant.
A composition comprising antigen-presenting cells, wherein the antigen-presenting cells comprise at least a first T. cruzi polypeptide antigen and a genetic adjuvant.
A method for treating or preventing an infection with an intracellular pathogen in a subject comprising administering to the subject an effective amount of antigen presenting cells, said cells comprising comprise at least a first polypeptide antigen from the intracellular pathogen and an expression vector encoding said first polypeptide antigen.
A method for preparing T. cruzi antigen presenting dendritic cells comprising:
- (a) obtaining immature antigen presenting cell precursors from a subject;
- (b) culturing the immature antigen presenting cell precursors to induce differentiation into immature dendritic cells;
- (c) transducing the immature dendritic cells with a vector for the expression of T. cruzi antigens;
- (d) contacting the immature dendritic cells with a recombinant T. cruzi antigen;
- and
- (e) culturing the immature dendritic cells to produce mature dendritic cells.
