Abstract
This invention relates to compositions and methods for modifying Soluble NSF Attachment Protein18 (SNAP18) genes in soybean plants to increase resistance to Soybean Cyst Nematode (SCN). The invention further relates to plants having increased resistance to/reduced severity of infection by SCN produced using the methods and compositions of the invention.
Claims28
A soybean plant or plant part thereof comprising at least one non-natural mutation in the C-terminal region of an endogenous Soluble NSF Attachment Protein18 (SNAP18) protein.

A method of generating variation in a C-terminal region of a soybean SNAP18 protein, comprising:
- introducing an editing system into a soybean plant cell, wherein the editing system is targeted to a region of an endogenous SNAP18 gene that encodes the C-terminal region of the SNAP18 protein;
- and
- contacting the region of the endogenous SNAP18 gene with the editing system, thereby introducing a mutation into the C-terminal region of the SNAP18 protein and generating variation in the C-terminal region of the soybean SNAP18 protein, optionally wherein generating variation in the C-terminal region of a soybean SNAP18 protein in a plant results in the plant having increased resistance to SCN or reduced severity of infection by SCN.
(canceled)
The method of claim 31, wherein contacting the region of the endogenous SNAP18 gene in the plant cell with the editing system produces a plant cell comprising in its genome an edited endogenous SNAP18 gene, the method further comprising (a) regenerating a plant from the plant cell;
- (b) selfing the plant to produce progeny plants (E1);
- (c) assaying the progeny plants of (b) for SCN resistance;
- and (d) selecting the progeny plants exhibiting increased SCN resistance or reduced severity of infection by SCN to produce selected progeny plants exhibiting increased SCN resistance or reduced severity of infection by SCN as compared to an unedited control plant.
38-44. (canceled)
A method for producing a soybean plant or part thereof comprising at least one cell having a mutated endogenous SNAP18 gene, the method comprising
- contacting a target site in an endogenous SNAP18 gene in the soybean plant or plant part with a nuclease comprising a cleavage domain and a DNA-binding domain, wherein the DNA binding domain binds to a target site in the endogenous SNAP18 gene,
- wherein the endogenous SNAP18 gene
- (a) encodes a sequence having at least 95% sequence identity to the amino acid sequence of SEQ ID NO:57;
- (b) comprises a sequence having at least 90% sequence identity to the nucleotide sequence of SEQ ID NO:55 or SEQ ID NO:56;
- and/or
- (c) comprises a sequence having at least 90% identity to any one of the nucleotide sequences of SEQ ID NO:61-64;
- to produce a plant cell comprising a mutation in the endogenous SNAP18 gene, thereby producing the soybean plant or part thereof comprising at least one cell having a mutation in the endogenous SNAP18 gene.
