Abstract
The field is related to plant breeding and methods of identifying and selecting plants with resistance to southern corn mst. Provided are methods to identify novel genes that encode proteins providing plant resistance to southern corn mst and uses thereof. These disease resistant genes are useful in the production of resistant plants through breeding, transgenic modification, or genome editing.

A method of identifying a plant with increased resistance to southern corn rust, said method comprising:
- a) screening a population of plants for the presence of a resistant gene allele that is associated with increased resistance to southern corn rust, wherein said resistant gene allele comprises an “A” at SNP001 (position 201 of reference sequence SEQ ID NO:
- 6), an “A” at SNP002 (position 127 of reference sequence SEQ ID NO:
- 7), a “C” at SNP003 (position 143 of reference sequence SEQ ID NO:
- 8), a “G” at S26 (position 43 of reference sequence SEQ ID NO:
- 1), an “A” at S27 (position 21 of reference sequence SEQ ID NO:
- 2), a “C” at S-24-1 (position 18 of reference sequence SEQ ID NO:
- 3), a “C” at S24-2 (position 19 of reference sequence SEQ ID NO:
- 3), a “G” at S24-3 (position 20 of reference sequence SEQ ID NO:
- 3), a “C” at S20-1 (position 17 of reference sequence SEQ ID NO:
- 4), a “T” at S20-2 (position 18 of reference sequence SEQ ID NO:
- 4), a “T” at S20-3 (position 21 of reference sequence SEQ ID NO:
- 4), a “C” at S28-1 (position 48 of reference sequence SEQ ID NO:
- 5), an “A” at S28-2 (position 49 of reference sequence SEQ ID NO:
- 5), a “T” at S28-3 (position 63 of reference sequence SEQ ID NO:
- 5), a “C” at S28-4 (position 64 of reference sequence SEQ ID NO:
- 5), an “A” at S28-5 (position 65 of reference sequence SEQ ID NO:
- 5), an “A” at S28-6 (position 66 of reference sequence SEQ ID NO:
- 5), a “G” at SNP004 (position 51 of reference sequence SEQ ID NO:
- 9), an “A” at SNP005 (position 39 of reference sequence SEQ ID NO:
- 10), or an “A” at SNP006 (position 23 of reference sequence SEQ ID NO:
- 11);
- and
- b) identifying plants having the allele associated with increased resistance to southern corn rust, wherein said plants have increased resistance to southern corn rust.
A method of increasing resistance to southern corn rust in a plant, comprising expressing in a plant a recombinant polynucleotide that encodes a polypeptide having an amino acid sequence of at least 90% sequence identity to the amino acid sequence set forth in SEQ ID NO:14;
- wherein the plant expressing the recombinant polypeptide has increased resistance to southern corn rust in the plant when compared to a control plant not comprising the recombinant polynucleotide.
A method of identifying a variant of the RppK gene that gives plants increased resistance to southern corn rust, the method comprising the steps of:
- a) generating variants of the RppK gene by combining through gene shuffling one or more nucleotide sequences encoding one or more fragments of SEQ ID NO:
- 14, or one or more nucleotides encoding one or more fragments of a protein that is at least 90% identical to SEQ ID NO:
- 14 and
- identifying a generated variant from said step a) that exhibits increased resistance to southern corn rust when expressed in a plant;
- or
- b) evaluating allelic variations in a population of plants that exhibit differing levels of southern corn rust resistance, wherein said plants are evaluated with respect to the polynucleotide sequence encoding a protein of SEQ ID NO:
- 14, or the genomic region that regulates the expression of the polynucleotide encoding the protein of SEQ ID NO:
- 14;
- and
- identifying an allelic variant from step b) that exhibits increased resistance to southern corn rust when expressed in a plant.
