Abstract
Compositions and methods for identifying antigen-specific T cells, including determining paired T cell receptor sequences for a specific antigen, are described. Compositions and methods for identifying neoantigen-specific T cells are also described.
Claims20
A method for isolating an antigen-specific T cell, the method comprising:
- a) contacting a sample with a particle comprising an MHC-antigen trimer and a DNA barcode trimer in conditions sufficient for a T cell to bind the particle;
- and
- b) isolating an antigen-specific T cell.
A method for identifying TCR sequences of an antigen-specific T cell, the method comprising:
- a) contacting a sample with a particle comprising an MHC-antigen trimer and a DNA barcode trimer in conditions sufficient for a T cell to bind the particle;
- b) isolating an antigen-specific T cell;
- and
- c) obtaining the TCR sequences.
