Abstract
K-Ras is the most frequently mutated oncogene in human cancer. Disclosed herein are compositions and methods for modulating K-Ras and treating cancer.

A method of treating cancer in a patient in need of thereof, the method comprising administering to the patient a therapeutically effective amount of a compound;
- wherein the compound is capable of covalently binding to cysteine residue 12 of a Ras protein and contacting a residue of a Ras protein Switch 2 binding pocket.
