Abstract
The present invention relates generally to growth of bacteria. More specifically the invention relates to a method for oxidative environment adaptation of anaerobic microorganisms and their use in developing new probiotics. In particular, the present invention provides a method for adaptation of anaerobic microorganisms and selection of more oxygen tolerant anaerobic microorganisms, said method comprising the steps of culturing said microorganisms with a stepwise dual induction of oxidative stress via applied voltage and oxygen diffusion, and a stepwise change of anti-oxidant/oxidized counterpart concentration ratio to adjust the redox state. New strains are also provided.
Claims19
An oxygen adapted strain of an anaerobic microorganism produced by
- (a) culturing anaerobic microorganisms under conditions comprising a stepwise dual induction of oxidative stress by (i) increasing in a stepwise manner an applied voltage and oxygen diffusion, and (ii) reducing in a stepwise manner the concentration of an antioxidant combined with increasing in a stepwise manner the concentration of an oxidized counterpart, thereby changing the anti-oxidant/oxidized counterpart concentration ratio and adjusting the redox state to produce an oxygen adapted anaerobic microorganism strain having increased oxygen tolerance;
- and
- (b) selecting the oxygen adapted anaerobic microorganism strain.
An oxygen adapted Faecalibacterium prausnitzii strain selected from the group consisting of DSM 32380, DSM 32378 and DSM 32379.
A method for enhancing production of oxygen adapted strains of anaerobic microorganisms, comprising
- (a) culturing anaerobic microorganisms under conditions comprising a stepwise dual induction of oxidative stress by (i) increasing in a stepwise manner an applied voltage and oxygen diffusion, and (ii) reducing in a stepwise manner the concentration of an antioxidant combined with increasing in a stepwise manner the concentration of an oxidized counterpart, thereby changing the anti-oxidant/oxidized counterpart concentration ratio and adjusting the redox state to produce oxygen adapted strains of anaerobic microorganisms;
- (b) selecting the oxygen adapted strains of anaerobic microorganisms to provide selected oxygen adapted strains of anaerobic microorganisms;
- and
- (c) further culturing the selected oxygen adapted strains of anaerobic microorganisms under constant conditions that correspond to final conditions reached at the end of the stepwise dual induction of (a) to enable a selective pressure on the oxygen adapted strains of anaerobic microorganisms, the final conditions comprising a combination of constant oxidizing potential/applied voltage, oxygen diffusion, and anti-oxidant/oxidized counterpart concentration ratio, whereby only strains that remain oxygen adapted survive, thereby enhancing production of oxygen adapted strains of anaerobic microorganisms.
