Abstract
Methods of testing tumor samples for mutational burden and/or for expression profiles permit the prediction of responsiveness of an individual to immunotherapy comprising PVSRIPO. Those predicted to respond are treated with PVSRIPO and those predicted not to respond are treated with other agents.
Claims25
A method of treating an individual having a tumor, comprising:
- testing a tumor sample from the individual and determining its tumor mutational burden, wherein (a) upon determining a high mutational burden in the tumor sample, administering to the individual a therapy that does not comprise PVSRIPO;
- or (b) upon determining a low mutational burden in the tumor sample, administering to the individual an immunotherapeutic agent comprising PVSRIPO.
A method for predicting response of an individual with cancer to treatment with an immunotherapeutic agent comprising PVSRIPO, the method comprising:
- testing a tumor sample from the individual and determining a high or a low tumor mutational burden;
- predicting that the individual is not likely to respond with a therapeutic effect to treatment with PVSRIPO when a high tumor mutational burden is detected in the tumor sample;
- and predicting that the individual is likely to respond with a therapeutic effect to treatment with PVSRIPO when a low tumor mutational burden is detected.
A method of determining if a treatment regimen for an individual with cancer should include treatment with PVSRIPO, the method comprising:
- testing a tumor sample from the individual with cancer and detecting tumor mutational burden;
- detecting high tumor mutational burden in the tumor sample;
- and
- determining that the treatment regimen will not include PVSRIPO and the individual will not be treated with.
A method of determining if a treatment regimen for an individual with cancer should include treatment with PVSRIPO, the method comprising:
- testing a tumor sample from the individual with cancer and detecting a low tumor mutational burden;
- and
- determining that the treatment regimen will include and the individual will be treated with PVSRIPO.
A method of treating an individual having a tumor, comprising:
- testing a tumor sample from the individual and determining expression of cytolytic activity genes, T-Cell Receptor (TCR) signaling genes, or inflammatory response genes, wherein (a) upon determining a low level of expression of cytolytic activity genes, T-Cell Receptor (TCR) signaling genes, or inflammatory response genes, in the tumor sample, administering to the individual a therapy that does not comprise PVSRIPO;
- and (b) upon determining a high level of expression of cytolytic activity genes, T-Cell Receptor (TCR) signaling genes, or inflammatory response genes in the tumor sample, administering to the individual an immunotherapeutic agent comprising PVSRIPO.
A method for predicting response of an individual with cancer to treatment with an immunotherapeutic agent comprising PVSRIPO, the method comprising:
- testing a tumor sample from the individual and determining a high or a low level of expression of cytolytic activity genes, T-Cell Receptor (TCR) signaling genes, or inflammatory response genes;
- predicting that the individual is not likely to respond with a therapeutic effect to treatment with PVSRIPO when a low level of expression of cytolytic activity genes, T-Cell Receptor (TCR) signaling genes, or inflammatory response genes is determined in the tumor sample;
- or
- predicting that the individual is likely to respond with a therapeutic effect to treatment with PVSRIPO when a high level of expression of cytolytic activity genes, T-Cell Receptor (TCR) signaling genes, or inflammatory response genes is determined.
A method of determining if a treatment regimen for an individual with cancer should include treatment with PVSRIPO, the method comprising:
- testing a tumor sample from the individual with cancer and detecting expression of cytolytic activity genes, T-Cell Receptor (TCR) signaling genes, or inflammatory response genes;
- detecting a low level of expression of cytolytic activity genes, T-Cell Receptor (TCR) signaling genes, or inflammatory response genes;
- and
- determining that the treatment regimen will not include PVSRIPO and the individual will not be treated with PVSRIPO.
A method of determining if a treatment regimen for an individual with cancer should include treatment with PVSRIPO, the method comprising:
- testing a tumor sample from the individual with cancer and detecting expression levels of cytolytic activity genes, T-Cell Receptor (TCR) signaling genes, or inflammatory response genes;
- detecting a high level of said expression;
- and
- determining that the treatment regimen will include and the individual will be treated with PVSRIPO.
