Abstract
Disclosed is an expression system for co-expressing multiple human cytomegalovirus (HCMV) antigens to stimulate potent humoral and cellular immune responses against HCMV infection. The expression system may include a vector inserted with multiple nucleic acid sequences that encode multiple subunits of HCMV antigens linked by one or more linking sequences such that the subunits are co-expressed simultaneously. Also disclosed are vaccine compositions comprising the expression system or the vector and methods of preventing or treating HCMV infections using the vaccine compositions.
Claims29
An expression system comprising one or more vectors, each vector comprising one or more expression cassettes, each expression cassette comprising:
- a single promoter;
- multiple nucleic acid sequences that encode multiple HCMV antigens,
- one or more 2A peptide signal sequences that mediate ribosomal skipping or one or more internal ribosomal entry sites between each of the multiple nucleic acid sequences;
- wherein the multiple HCMV antigens, when co-expressed by the expression system simultaneously, effectively elicit multi-functional humoral and cellular immune responses.
An expression system comprising a recombinant bacterial artificial chromosome (BAC) construct comprising a BAC vector and a viral vector genome, the viral vector genome comprising at least one expression cassette, wherein each expression cassette comprises:
- a single promoter;
- two or more nucleic acid sequences that encode two or more HCMV antigens;
- one or more 2A signal sequences between each of the two or more nucleic acid sequences;
- wherein the two or more HCMV antigens are simultaneously expressed by the expression system.
A vaccine composition for preventing or treating HCMV infection comprising a viral vector capable of co-expressing two, three, four, five, six, seven, eight, or more HCMV antigens simultaneously;
- and a pharmaceutically acceptable carrier, adjuvant, additive or combination thereof.
A method of preventing HCMV entry into a cell comprising infecting the cell with an effective amount of a viral vector, the viral vector capable of co-expressing seven or more HCMV antigens simultaneously.
A method of preventing or treating HCMV infection in a subject comprising administering a prophylactically or therapeutically effective amount of a vaccine to the subject, wherein the vaccine comprises a viral vector capable of co-expressing seven or more HCMV antigens simultaneously, and a pharmaceutically acceptable carrier, adjuvant, additive or combination thereof.
