## Abstract

Herein is reported a multimeric fusion polypeptide comprising five monomeric fusion polypeptides each comprising at least a Fab fragment and a COMP-domain of SEQ ID NO: 01 or a functional fragment thereof.

## Claims

### Claim 1

A multimeric, multispecific fusion polypeptide
comprising five monomeric fusion polypeptides each comprising at least a Fab and a COMP-domain of SEQ ID NO: 01 or a fragment of the COMP-domain of SEQ ID NO: 01 that can associate with other COMP-domains.

### Claim 2 (depends on 1)

The multimeric fusion polypeptide according to claim 1, wherein the Fab is conjugated either directly or via a peptidic linker to the COMP-domain independently of each other for the five monomeric fusion polypeptides.

### Claim 3 (depends on 1, 2)

The multimeric fusion polypeptide according to any one of claims 1 to 2, wherein the COMP-domain is conjugated to a C-terminus or an N-terminus of the Fab independently of each other for the five monomeric fusion polypeptides.

### Claim 4 (depends on 1, 2, 3)

The multimeric fusion polypeptide according to any one of claims 1 to 3, wherein the Fab is selected from a Fab, a scFab, a bispecific Fab and a DAF.

### Claim 5 (depends on 1, 2, 3)

The multimeric fusion polypeptide according to any one of claims 1 to 3, wherein the Fab is a half-antibody.

### Claim 6 (depends on 5)

The multimeric fusion polypeptide according to claim 5, wherein the half-antibody is a dimeric half antibody wherein one monomer comprises the hole mutations and the respective other monomer comprises the knob mutation.

### Claim 7 (depends on 6)

The multimeric fusion polypeptide according to claim 6, wherein the first half antibody binds to a first antigen and the second half antibody binds to a second antigen (bispecific antibody).

### Claim 8 (depends on 1, 2, 3, 4, 5, 6, 7)

A method for the preparation of the multimeric fusion polypeptide according to any one of claims 1 to 7, comprising the steps of
transforming a host cell with expression vectors comprising nucleic acids encoding the multimeric fusion polypeptide,
culturing said host cell under conditions that allow synthesis of said multimeric fusion polypeptide, and
recovering said multimeric fusion polypeptide from said host cell culture.

### Claim 9 (depends on 8)

A multimeric fusion polypeptide produced by the method according to claim 8.

### Claim 10 (depends on 1, 2, 3, 4, 5, 6, 7)

A nucleic acid encoding the multimeric fusion polypeptide according to any one of claims 1 to 7.

### Claim 11 (depends on 10)

An expression vector comprising a nucleic acid according to claim 10.

### Claim 12 (depends on 10)

A host cell comprising a nucleic acid according to claim 10.

### Claim 13 (depends on 1, 2, 3, 4, 5, 6, 7)

A pharmaceutical composition comprising the multimeric fusion polypeptide according to any one of claims 1 to 7 in combination with at least one pharmaceutically acceptable carrier.
