Abstract
The present invention relates to methods of upgrading biomass to provide useful chemical intermediates, fuels, amino acids, nutrients, etc. In particular examples, the biomass is a by-product of ethanol production and is mainly used as high-protein feed. Described herein are methods for upgrading such biomass, such as by implementing pre-treatment conditions and by employing fermentation conditions including modified organisms.
Claims10
An isolated, genetically engineered organism comprising:
- a modified alcohol dehydrogenase having increased reactivity with nicotinamide adenine dinucleotide (NADH), as compared to a wild-type alcohol dehydrogenase;
- and
- a modified ketol-acid reductoisomerase having increased reactivity with NADH, as compared to a wild-type ketol-acid reductoisomerase.
An isolated nucleic acid comprising a nucleic acid sequence having at least 90% sequence identity to any one of SEQ ID NOs:14-25 or a complement of any of these, or a fragment thereof,
- wherein the isolated nucleic acid encodes a polypeptide sequence having at least 90% sequence identity to any one of SEQ ID NOs:1-5, or a fragment thereof, and
- wherein the isolated nucleic acid encodes a modified alcohol dehydrogenase having increased reactivity with nicotinamide adenine dinucleotide (NADH), as compared to a wild-type alcohol dehydrogenase.
A vector comprising:
- a first nucleic acid sequence encoding a modified alcohol dehydrogenase, wherein the first nucleic acid sequence has at least 90% sequence identity to any one of SEQ ID NOs:14-25 or a complement of any of these, or a fragment thereof;
- and
- a second nucleic acid sequence encoding a modified ketol-acid reductoisomerase, wherein the second nucleic acid has at least 90% sequence identity to any one of SEQ ID NOs:26-31 or a complement of any of these, or a fragment thereof.
