Abstract
Disclosed herein are methods for generating SC-β cells using chemically defined, completely serum free media, and isolated populations of SC-β cells for use in various applications, such as cell therapy.
Claims30
A serum free in vitro composition that comprises a ROCK inhibitor and a plurality of foregut endoderm cells that express FOXA2 and SOX2.
A method of in vitro cell differentiation, comprising:
- contacting an in vitro cell population comprising foregut endoderm cells that express FOXA2 and SOX2 with a ROCK inhibitor, a PKC activator, an FGF family growth factor, a sonic hedgehog pathway inhibitor, and a retinoic acid signaling pathway activator;
- wherein the cell population is in serum-free media, and wherein the method results in a plurality of the foregut endoderm cells in the population to differentiate into pancreatic progenitor cells that express PDX1.
