An isolated anti-PD-1 construct comprising a single-domain antibody (sdAb) moiety specifically recognizing PD-1, wherein the sdAb moiety comprises a CDR1 comprising the amino acid sequence of any one of SEQ ID NOs:
- 37-68 and 70, or a variant thereof comprising up to about 3 amino acid substitutions;
- a CDR2 comprising the amino acid sequence of any one of SEQ ID NOs:
- 109-140 and 142, or a variant thereof comprising up to about 3 amino acid substitutions;
- and a CDR3 comprising the amino acid sequence of any one of SEQ ID NOs:
- 181-212 and 214, or a variant thereof comprising up to about 3 amino acid substitutions.
