Abstract
The present invention relates inter alia to fertile non-human vertebrates such as mice and rats useful for producing antibodies bearing human variable regions, in which endogenous antibody chain expression has been inactivated.
Claims21
A recombinant rodent whose genome comprises immunoglobulin heavy chain (IgH) locus-containing chromosome, said chromosome comprising a transgenic IgH locus functional to express a chimeric IgH polypeptide and non-functional to express endogenous IgH polypeptide, wherein said transgenic IgH locus comprises one or more human VH gene segments, one or more human D gene segments and one or more human JH gene segments positioned 5′ to a rodent or human heavy chain constant (CH) region;
- wherein said chromosome comprises a deletion of all or part of the rodent endogenous heavy chain VDJ region, including deletion of endogenous ADAM6-encoding nucleotide sequences;
- and wherein said genome comprises integrated exogenous ADAM6-encoding nucleotide sequence functional to express ADAM6 protein.
A recombinant non-human vertebrate or recombinant non-human vertebrate cell having a recombinant genome comprising a transgenic immunoglobulin (Ig) locus functional to express chimeric Ig comprising a human Ig variable region, and wherein said genome comprises V region coding sequences endogenous to said non-human vertebrate or said non-human vertebrate cell, wherein said endogenous Ig V region coding sequences are positioned in said recombinant genome on a chromosome that does not comprises endogenous Ig V region coding sequences in a corresponding position in a non-recombinant (native) genome, and wherein said recombinant genome is non-functional to express endogenous Ig.
