Abstract
Disclosed are methods for treating or preventing or delaying outset of Alzheimer's disease (AD) in a subject by targeting the novel pathway STAT1-CH25H in AD pathogenesis, specifically by administering to the subject a pharmaceutically effective amount of a STAT1 inhibitor, a CH25H inhibitor, or a 25-OHC inhibitor, for example, a 3-hydroxy-3-methyl-glutaryl-coenzyme A (HMG-CoA) reductase inhibitor such as simvastatin.
Claims23
A method of treating Alzheimer's disease (AD) in a subject, the method comprising administering to the subject a pharmaceutically effective amount of a STAT1 inhibitor, a CH25H inhibitor, and/or a 25-OHC inhibitor.

A method of preventing or delaying onset of Alzheimer's disease (AD) in a subject, the method comprising administering to the subject a pharmaceutically effective amount of a STAT1 inhibitor, a CH25H inhibitor, and/or a 25-OHC inhibitor.
22-40. (canceled)
A method of treating or preventing or delaying the onset of Alzheimer's disease (AD) in a subject, the method comprising the step of:
- a. providing a genome editing tool;
- b. delivering the genome editing tool to neural cells;
- and
- c. editing the STAT1 gene by deleting the entire STAT1 gene, the phosphorylation site of the STAT1 gene, the promoter region of the STAT1 gene, or the SH2 domain of the STAT1 gene, and/or editing the CH25H gene by deleting the entire CH25H gene, the histone cluster regions of the CH25H gene, or the promoter region of the CH25H gene.
