Abstract
The present invention relates to a cell line for producing an adenovirus having a limited autoreplication capability, and a method of preparing the same, and more particularly, to a cell line for producing an adenovirus having no autoreplication capability by expressing at least one selected from an adenoviral E1 protein and an E1A or E1B protein, and a method of preparing the same. Also, the present invention relates to the use of the cell line expressing at least one selected from the adenoviral E1 protein and the E1A or E1B protein.
Claims13
A cell line for producing an adenovirus having a limited autoreplication capability, comprising:
- at least one of an E1 protein and a nucleic acid sequence encoding the same;
- and
- at least one of an E1A protein, a nucleic acid sequence encoding the E1A protein, an E1B protein, and a nucleic acid sequence encoding the E1B protein.
A method of preparing a cell line for producing an adenovirus having a limited autoreplication capability, the method comprising:
- (a) preparing each retrovirus vector comprising a nucleic acid sequence of the adenovirus E1, E1A and E1B genes, respectively;
- (b) transfecting the each retrovirus vector into a cell line for producing a retrovirus to produce a retrovirus, respectively;
- (c) infecting a cell line for producing an adenovirus with the retrovirus comprising the nucleic acid sequence of the E1 gene, which is obtained from the (b);
- and
- (d) further infecting at least one of the retroviruses comprising the nucleic acid sequences of E1A or E1B gene, which are obtained from the (b) into the cell line for producing an adenovirus of (c),
- to obtain the cell line for producing the adenovirus having a limited autoreplication capability.
A packaging system for AAV (adeno-associated virus) production comprising:
- an adenovirus obtained from a cell line for producing an adenovirus having a limited autoreplication capability, wherein the cell line comprises:
- at least one of an E1 protein and a nucleic acid sequence encoding the same;
- and
- at least one of E1A protein, nucleic acid sequences encoding E1A, E1B protein and nucleic acid sequences encoding E1B;
- a cell line comprising a nucleic acid sequence of a rep gene of the AAV and a nucleic acid sequence of a cap gene of the AAV;
- and
- an AAV vector.
A composition for gene therapy comprising:
- an adenovirus obtained from a cell line for producing an adenovirus having a limited autoreplication capability, wherein the cell line comprises:
- at least one of E1 protein and the nucleic acid sequences encoding the same and;
- and
- at least one of E1A protein, nucleic acid sequences encoding E1A, E1B protein and nucleic acid sequences encoding E1B.
