Abstract
This invention relates to agents that are inhibitors or activators of variant forms of endogenous proteins and novel methods of identifying such variants. Of particular interest are inhibitors and activators of endogenous protein variants, encoded by genes which have mutated, which variants often arise or are at least first identified as having arisen following exposure to a chemical agent which is known to be an inhibitor or activator of the corresponding unmutated endogenous protein.

A method of testing a compound, the method comprising:
- a) incubating a test cell which expresses a theramutein with a compound, the theramutein being capable of producing a detectable phenoresponse of the test cell;
- b) incubating a control cell which expresses a prototheramutein corresponding to the theramutein with a known modulator of the prototheramutein, the prototheramutein being capable of producing the phenoresponse in the control cell;
- c) comparing the phenoresponse of the control cell to the known modulator of the prototheramutein to the phenoresponse of the test cell to the compound;
- and
- d) determining that the compound is a modulator of the theramutein when the phenoresponse of the test cell is modulated to at least the same degree as the phenoresponse of the control cell is modulated by the known modulator of the prototheramutein.
A method of testing the ability of a compound to inhibit or activate a selected theramutein in a cell, the method comprising:
- a) providing a test cell that expresses a theramutein, wherein the test cell is capable of producing a detectable phenoresponse;
- b) providing a control cell which expresses a prototheramutein corresponding to the theramutein, wherein the control cell is capable of producing the phenoresponse;
- c) treating the test cell with a compound;
- d) treating the control cell with the known modulator;
- e) comparing the phenoresponse of the control cell to the known modulator of the prototheramutein to the phenoresponse of the test cell to the compound;
- and
- f) determining that the compound inhibits or activates the theramutein when the phenoresponse of the test cell is modulated to at least the same degree as the phenoresponse of the control cell is modulated by the known modulator of the prototheramutein.
