Abstract
Disclosed are compounds of formula (I) below and pharmaceutically acceptable salts thereof: (I), in which each of variables R1, R2, R3, R4, R5, A1, A2, A3, A4, X and Y is defined herein. Also disclosed are methods for reducing the glycemic level and treating glucagon-associated disorders with a compound of formula (I) or a salt thereof and a pharmaceutical composition containing same.

A compound of formula (I):
- or a pharmaceutically acceptable salt thereof,
- wherein
- each of A1, A2, A3 and A4 independently is CR6 or N, with the proviso that no more than two of A1, A2, A3, and A4 are N;
- X is —CH2—, —NH—, —O—, —S— or a direct bond;
- Y is —(C═O)—, —CH2— or a direct bond;
- R1 is C1-6 alkyl, C1-6 alkoxy, aryl, heteroaryl, C3-12 cycloalkyl, C1-12 heterocycloalkyl, or —(C1-6 alkyl)-(C3-12 cycloalkyl);
- the C1-6 alkyl being optionally substituted with one to three moieties selected from the group consisting of halo, hydroxyl, C1-6 alkoxy, aryl and heteroaryl;
- each of the C1-6 alkoxy, aryl, heteroaryl, C3-12 cycloalkyl, and C1-12 heterocycloalkyl being optionally substituted with one to three moieties selected from the group consisting of halo, aryl, C1-6 alkyl optionally substituted with one to three halo and C1-6 alkoxy optionally substituted with one to three halo;
- the heteroaryl being optionally fused with one aryl moiety;
- and the aryl being optionally substituted with heteroaryl or heteroaryl fused with one aryl moiety;
- R2 is C1-6 alkyl,
- R3 is H, methyl or ethyl;
- R4 is H, C1-6 alkyl, or C1-6 alkoxy, each of the C1-6 alkyl and C1-6 alkoxy being optionally substituted with one to three halo;
- R5 is tetrazolyl, —CH2-tetrazolyl, —CH2CH2CO2R10, —CH2CH(OH)CO2H, or —(CH2)2SO3H;
- R6 is H, halo, C1-6 alkyl, or C1-6 alkoxy, each of the C1-6 alkyl and C1-6 alkoxy being optionally substituted with one to three halo;
- R7 is H, halo, C1-6 alkyl or C1-6 alkoxy;
- R8 is H, halo, C1-6 alkyl or C1-6 alkoxy;
- R9 is H, halo, C1-6 alkyl, C1-6 alkoxy, C3-12 cycloalkyl, aryl or heteroaryl;
- the C1-6 alkyl being optionally substituted with one to three halo;
- each of the C1-6 alkoxy, C3-12 cycloalkyl, aryl and heteroaryl being optionally substituted with one to three moiety selected from the group consisting of halo, aryl, C1-6 alkyl optionally substituted with one to three halo, C1-6 alkoxy optionally substituted with one to three halo, amino, and CN;
- and the heteroaryl being optionally fused with one aryl moiety or one heteroaryl moiety;
- R10 is H or C1-6 alkyl;
- and
- each of J1, J2, J3, J4 independently is C or N, with the proviso that no more than two of J1, J2, J3 and J4 are N.
