Abstract
The present invention relates to a brain tumor animal model that directly reflects the phenomenon in a human patient and a method of preparing the same, and more specifically, a brain tumor animal model that mutations are introduced into p53, Pten, and EGFR genes, a screening method of a therapeutic agent for a brain tumor using the animal model, and a preparing method thereof.
Claims15
A transgenic animal with glioma in which p53 and Pten genes are knocked out specifically in nerve stem cell of subventricular zone (SVZ) of Epidermal growth factor receptor (EGFR) knockout animal, wherein the nerve stem cell has normal cytoarchitecture and the glioma occurs in the cortical region separated from the SVZ, and is developed from Glial fibrillary acidic protein (GFAP)-positive nerve stem cell.
A method of preparing a transgenic animal having glioma in the cortical region, comprising the steps:
- (a) preparing an epidermal growth factor receptor (EGFR) knockout animal;
- (b) preparing a vector knocking out p53 and Pten genes;
- and
- (c) injecting the vector prepared in step (b) into the subventricular zone (SVZ) of the animal prepared in step (a) by electroporation.
