Abstract
Compositions, vaccines and methods using a combination of adenovirus vectors and MVA vectors for inducing protective immunity against a Marburg virus infection are described.
Claims21
A vaccine combination comprising
- a. a first composition comprising an immunologically effective amount of an adenovirus vector comprising a nucleic acid encoding a first antigenic protein of a Marburg virus, together with a pharmaceutically acceptable carrier;
- and
- b. a second composition comprising an immunologically effective amount of an MVA vector comprising a nucleic acid encoding a second antigenic protein of a Marburg virus, preferably the nucleic acid further encoding one or more antigenic proteins of at least one additional filovirus subtype, together with a pharmaceutically acceptable carrier;
- wherein the first antigenic protein and the second antigenic protein share at least one antigenic determinant, and the first composition is the priming composition and the second composition is the boosting composition.

A method of inducing an immune response against a Marburg virus in a subject, the method comprising:
- a. administering to the subject a first composition comprising an immunologically effective amount of an adenovirus vector comprising a nucleic acid encoding a first antigenic protein of a Marburg virus;
- and
- b. administering to the subject a second composition comprising an immunologically effective amount of an MVA vector comprising a nucleic acid encoding a second antigenic protein of a Marburg virus, preferably the nucleic acid further encoding one or more antigenic proteins of at least one additional filovirus subtype.
- wherein the first antigenic protein and the second antigenic protein share at least one antigenic determinant, and step (b) is conducted 1-12 weeks, preferably 4 or 8 weeks, after step (a).

A kit comprising:
- a. a first composition comprising an immunologically effective amount of an adenovirus vector comprising a nucleic acid encoding a first antigenic protein of a Marburg virus, together with a pharmaceutically acceptable carrier;
- and
- b. a second composition comprising an immunologically effective amount of an MVA vector comprising a nucleic acid encoding a second antigenic protein of a Marburg virus, preferably the nucleic acid further encoding one or more antigenic proteins of at least one additional filovirus subtype, together with a pharmaceutically acceptable carrier;
- wherein the first antigenic protein and the second antigenic protein share at least one antigenic determinant, and the first composition is the priming composition and the second composition is the boosting composition.
