Abstract
The present invention relates to an in vitro method for diagnosing lupus in a subject, said method comprising the step of detecting in a biological sample obtained from the subject the autoantibody recognizing the protein biomarker THEX1. More, the invention relates to kits and array useful for carrying out diagnosis methods according to the present invention.

An analytical method for detecting auto-antibodies to Three prime Histone mRNA EXonuclease 1 (THEX1) in a subject, comprising
- obtaining a biological sample from the subject, wherein the biological sample does not have one or both of antitopoisomerase autoantibodies (ATAs) and anticentromere autoantibodies (ACAs);
- and
- detecting whether auto-antibodies to THEX1 are present in the biological sample.
A method of diagnosing and treating lupus in a subject, said method comprising the steps of
- obtaining a biological sample from the subject, wherein the subject does not have one or both of antitopoisomerase autoantibodies (ATAs) or anticentromere autoantibodies (ACAs);
- detecting whether auto-antibodies to Three prime Histone mRNA EXonuclease 1 (THEX1) are present in the sample, wherein detection of auto-antibodies to THEX1 indicates that the subject has lupus, and
- administering corticoids and/or hydroxychloroquine to a subject whose measurement is indicative of having lupus.
