Abstract
The present disclosure relates to the field of plant molecular biology, more particularly to regulation of gene expression in plants.
Claims40
A hybrid regulatory element comprising an assembly of heterologous regulatory element segments, wherein the assembly of the heterologous regulatory element segments produces a unique tissue expression pattern.
A hybrid regulatory element comprising at least one polynucleotide sequence selected from the group comprising:
- (a) a polynucleotide having at least 85 percent sequence identity to any one of the nucleic acid sequences of SEQ ID NOs:
- 11-30 and 35-60;
- (b) the polynucleotide of any one of SEQ ID NOs:
- 11-30 and 35-60;
- or
- (c) a fragment of SEQ ID NO:
- 11-30 and 35-60;
- wherein said polynucleotide sequence initiates transcription in a plant cell.
A method of creating a hybrid regulatory element comprising:
- (a) Identifying a native regulatory element with a regional expression pattern;
- (b) Identifying motifs of the native regulatory element;
- and
- (c) Assembling at least two segments into a hybrid regulatory element that initiates transcription in a plant cell.
A method for directed expression of a polynucleotide sequence in a plant or plant cell, said method comprising introducing into the plant or the plant cell an expression cassette comprising a hybrid regulatory element operably linked to a heterologous polynucleotide sequence of interest, wherein said hybrid regulatory element produces a unique tissue expression pattern of the heterologous polynucleotide sequence of interest.
