## Abstract

Methods and compounds are provided to improve the desorption and ionization of analyte for mass spectrometry analysis. More specifically, it is for electrospray ionization (ESI) mass spectrometry. The method uses charged affinity molecules that can bind with analyte either temporarily or permanently to improve the desorption and ionization of analyte. The charged affinity molecules can be positively charged or negatively charged.

## Claims

### Claim 1

A method to improve the detection of non-ionizable analyte molecule in electrospray ionization (ESI) mass spectrometry, comprising:
providing permanently charged affinity molecule having affinity moiety selected from cyclodextrin , calixarene, cucurbituril and pillararene that can bind with said analyte molecule via non-covalent bond;
mixing said charged affinity molecule with a sample solution containing said analyte to form a solution containing a non-covalently bound complex between said analyte molecule and said charged affinity molecule; and
performing electrospray ionization (ESI) mass spectrometry for the solution containing the bound complex , and
detecting the presence of the analyte molecule by detecting the presence of the bound complex in ESI mass spectrometry.

### Claim 2 (depends on 1)

The method according to claim 1, wherein the charged affinity molecule is positively charged.

### Claim 3 (depends on 1)

The method according to claim 1, wherein the charged affinity molecule is negatively charged.

### Claim 4 (depends on 1)

The method according to claim 1, wherein the charged affinity molecule contains quaternary ammonium cation group.

### Claim 5 (depends on 1)

The method according to claim 1, wherein the charged affinity molecule contains sulfate anion group.

### Claim 6

A compound to improve the signal of non-ionizable analyte molecule in electrospray ionization (ESI) mass spectrometry comprising a permanently charged motif and a binding motif selected from cyclodextrin, calixarene, cucurbituril and pillararene that can bind with the said analyte molecule to form a non-covalent complex.

### Claim 7 (depends on 6)

The compound according to claim 6, wherein the charged motif is a positively charged group.

### Claim 8 (depends on 6)

The compound according to claim 6, wherein the charged motif is negatively charged group.

### Claim 9 (depends on 6)

The compound according to claim 6, wherein the charged motif is a quaternary ammonium cation group.

### Claim 10 (depends on 6)

The compound according to claim 6, wherein the charged motif is sulfate anion group.
