Provided are a recombinant polynucleotide encoding a polypeptide including a reporter moiety, a substrate moiety, and a destabilization moiety, a host cell including the same, and use thereof to measure the level of a protease by using the recombinant polynucleotide.

A recombinant polynucleotide comprising a first polynucleotide that encodes a polypeptide that includes:
- a reporter moiety;
- a destabilization moiety;
- and a substrate moiety operatively linking the reporter moiety at an N-terminus to the destabilization moiety at a C-terminus, wherein the substrate moiety comprises a cleavage site of protease activity.
