Abstract
A cyclic polypeptide, derivative or analogue thereof, comprising an amino acid sequence derived from the C-terminus of acetylcholinesterase (ACh E), or a truncation thereof.
Claims23
1-3. (canceled)
A cyclic polypeptide of SEQ ID NO:
- 4, or a derivative or analogue thereof having at least 65% sequence identity to SEQ ID NO:
- 4.
5-8. (canceled)
A cyclic polypeptide, derivative or analogue thereof according to claim 1, wherein the cyclic polypeptide, derivative or analogue thereof comprises an amino acid sequence derived from the C-terminus of tailed acetylcholinesterase (T-AChE), or a truncation thereof.
10-29. (canceled)
A method of using the cyclic polypeptide, derivative or analogue thereof according to claim 1, in an in vitro or ex vivo analytical method for investigating the allosteric site of α7 nicotinic-receptor comprising the step of inhibiting additional influx of calcium through the α7 nicotinic-receptor.
(canceled)
The cyclic polypeptide, derivative or analogue thereof according to claim 1, having at least 75%, 85%, 95% or 99% sequence identity to SEQ ID No:
- 4.
The cyclic polypeptide, derivative or analogue thereof according to claim 1, wherein the polypeptide, derivative or analogue thereof is the cyclic polypeptide of SEQ ID No:
- 4.
The cyclic polypeptide, derivative or analogue thereof according to claim 1, wherein the cyclic polypeptide, derivative or analogue thereof is a selective allosteric modulator of the α7 nicotinic-receptor.
The cyclic polypeptide, derivative or analogue thereof according to claim 1, wherein the cyclic polypeptide, derivative or analogue thereof is a selective allosteric modulator of the α7 nicotinic-receptor, which prevents the additional influx of calcium through an allosteric site of the α7 nicotinic-receptor, and outcompetes binding for β-amyloid.
A method of treating, ameliorating or preventing a neurodegenerative disorder in a subject, the method comprising, administering to a subject in need of such treatment, a therapeutically effective amount of the cyclic polypeptide, derivative or analogue thereof according to claim 1.
A pharmaceutical composition comprising a therapeutically effective amount of a cyclic polypeptide, derivative or analogue thereof according to claim 1, and a pharmaceutically acceptable vehicle.
A process for making the pharmaceutical composition, comprising combining a therapeutically effective amount of a cyclic polypeptide, derivative or analogue thereof according to claim 1 with a pharmaceutically acceptable vehicle.
A receptor allosteric modulator comprising a cyclic polypeptide, derivative or analogue thereof according to claim 1.
