Abstract
The present invention relates to novel compounds of formula (I) and methods for the manufacture of inhibitors of deubiquitylating enzymes (DUBs). In particular, the invention relates to the inhibition of ubiquitin C-terminal hydrolase 30 or ubiquitin specific peptidase 30 (USP30). The invention further relates to the use of DUB inhibitors in the treatment of conditions involving mitochondrial dysfunction and in the treatment of cancer.
Claims25
A compound with the formula I:
- or a pharmaceutically acceptable salt thereof, wherein:
- R1b, R1c, R1d and R1e each independently represent hydrogen or an optionally substituted C1-C6 alkyl, or R1b is linked to R1c or R1a to form an optionally substituted cycloalkyl ring, or R1c is linked to R1d or R1f to form an optionally substituted cycloalkyl ring;
- R1a, R1f and R1g each independently represent hydrogen, fluorine, cyano, hydroxyl, amino, optionally substituted C1-C6 alkyl or optionally substituted C1-C6 alkoxy, or R1a is linked to R1b to form an optionally substituted cycloalkyl ring, or R1a or R1g is linked to R to form an optionally further substituted ring, or R1f is linked to R1e or R1g to form an optionally substituted cycloalkyl ring;
- R2 represents optionally substituted C1-C3 alkyl or together with R3 forms an optionally substituted 5 to 10 membered monocyclic or bicyclic ring, or R2 together with R1g or R1a forms an optionally further substituted 5 or 6 membered ring;
- R3 represents an optionally substituted 5 to 10 membered monocyclic or bicyclic heteroaryl or aryl ring or R3 together with R2 forms an optionally substituted 5 to 10 membered monocyclic or bicyclic ring;
- L represents a covalent bond or an optionally substituted C1-C3 alkylene.
