Abstract
Provided is a composition containing an agent capable of inhibiting the expression of HMGCLL1 IS3, and a method for treating or preventing a malignant tumor. The malignant tumor expresses HMGCLL1 IS3. The malignant tumors may show resistance to tyrosine kinase inhibitors. A method, composition, and kit for determining the expression level of HMGCLL1 IS3, for diagnosing drug resistance, for determining the probability of developing a malignant tumor, and for predicting prognosis of malignant tumors are provided.
Claims25
A composition comprising an agent capable of inhibiting an expression of 3-hydroxymethyl-3-methylglutaryl-CoA lyase like 1 isoform 3 (HMGCLL1 IS3).

A method selected from the following (i)-(ii):
- (i) a method for determining tyrosine kinase inhibitor resistance in a test subject, and
- (ii) a method for predicting prognosis of a malignant tumor in a test subject, said method comprising:
- (a) measuring an expression level of HMGCLL1 IS3 from a biological sample of the test subject who has a malignant tumor or is suspected to have a malignant tumor;
- and
- (b) comparing the measured expression level of HMGCLL1 IS3 of step (a) with that of a control sample of a control subject,
- wherein a higher expression level of HMGCLL1 IS3 measured in (a) than that of the control sample is an indicator of tyrosine kinase inhibitor resistance of the test subject or a poor prognosis of a malignant tumor of the test subject.
A composition comprising an agent capable of measuring the expression level of HMGCLL1 IS3.

A method selected from the following (i)-(ii):
- (i) a method for determining tyrosine kinase inhibitor resistance in a test subject, and
- (ii) a method for predicting prognosis of a malignant tumor in a test subject, said method comprising
- detecting or amplifying, from a biological sample isolated from the subject, a polynucleotide consisting of 2 to 51 continuous nucleotides comprising single nucleotide polymorphism (SNP) located at the 26th nucleotide in a polynucleotide consisting of nucleotides selected from the group consisting of nucleotide sequences of SEQ ID NOS:
- 9 to 16 and 33 to 173;
- or a complementary polynucleotide thereof,
- wherein a detection or amplification of the polynucleotide is indicative of tyrosine kinase inhibitor resistance of the test subject or a poor prognosis of a malignant tumor of the test subject.
