Abstract
Certain aspects of the present invention provide devices and methods for preparing oligonucleotides and for assembling nucleic acid molecules using microfluidic devices.
Claims15
A method for assembling a target polynucleotide using a microfluidic system, comprising:
- flowing a continuous carrier phase fluid on a solid support, wherein the continuous carrier phase fluid comprises one or more droplets in fluid communication with the solid support, wherein the one or more droplets are immiscible with the carrier phase fluid, wherein the solid support comprises a plurality of features comprising oligonucleotides that together comprise the target polynucleotide;
- and
- displacing the one or more droplets from a first feature to a second feature to allow assembly of the oligonucleotides into the target polynucleotide.
