Abstract
The present disclosure relates to CRISPR-Cas10 systems and methods for phage genome editing.
Claims20
A phage genome editing system comprising:
- a Staphylococcus bacterial cell that can be infected by a phage;
- a vector comprising a crRNA that can hybridize to a nucleic acid sequence of the phage;
- and
- a vector comprising a donor nucleic acid sequence, wherein the donor nucleic acid sequence comprises:
- a mutated nucleic acid sequence to be introduced into the phage, flanked by two nucleic acid sequences containing regions of homology to the phage genome.
2 more elements
A method for editing a phage genome, comprising:
- introducing into a Staphylococcus bacterial cell a vector comprising:
- a crRNA that can hybridize to a nucleic acid sequence of the phage and a donor nucleic acid sequence, wherein the donor nucleic acid sequence comprises:
- a mutated nucleic acid sequence to be introduced into the phage, flanked by two nucleic acid sequences containing regions of homology to the phage genome;
- introducing a phage into the cell;
- and
- editing the phage genome to incorporate the mutated nucleic acid sequence.
3 more elements
A phage genome editing system comprising:
- a Staphylococcus bacterial cell that can be infected by a phage;
- a vector comprising a crRNA that can hybridize to a nucleic acid sequence of the phage;
- a vector comprising a CRISPR-Cas10 system, wherein the CRISPR-Cas10 system comprises CRISPR-associated genes csm1/cas10, csm2, csm3, csm4, csm5, csm6, and/or cas6;
- and
- a vector comprising a donor nucleic acid sequence, wherein the donor nucleic acid sequence comprises:
- a mutated nucleic acid sequence to be introduced into the phage, flanked by two nucleic acid sequences containing regions of homology to the phage genome.
