Abstract
Disposable fluid circuits, systems utilizing disposable fluid circuits and methods for combining a treating agent with separated blood component are disclosed. The treating agent is contained within a container and the disposable fluid circuit includes one or more frangible cannulas located in one or more flow paths of the circuit that are selectively opened to establish flow communication between the treating agent container and a treatment container, and/or a separation chamber or other container.
Claims20
A disposable fluid circuit for combined association with a cell separator and a separate irradiation device comprising:
- a) a patient access device for establishing flow communication with a patient;
- b) a cell separation chamber in openable fluid communication with said patient access device;
- c) a treatment container comprising walls made of a material that is transparent to light of a selected wavelength;
- d) tubing defining a sterile flow path between said separation chamber and said treatment container, said flow path having length sufficient to allow for placement of said separation chamber within said separator and allow for placement of said treatment container is said separate irradiation device;
- e) a container of photoactive agent integral with said circuit, said container having an outlet in openable flow communication with an outlet flow path;
- and
- f) a frangible cannula for preventing flow of photoactive agent from said container of photoactive agent to said treatment container.
A system for performing a therapeutic cellular treatment comprising:
- a) a reusable separator unit for separating blood into blood components;
- b) a reusable irradiation unit for treating a separated blood component;
- c) a controller configured to effect the separation of blood into blood components, effect the conveying of the separated blood components through a disposable fluid circuit and, optionally, to effect the treatment of said separated component;
- d) a disposable fluid circuit for combined association with a cell separator and a separate irradiation device comprising:
- i. a patient access device for establishing flow communication with a patient;
- ii. a cell separator comprising a separation chamber in openable fluid communication with said patient access device;
- iii. a treatment container comprising walls made of a material that is transparent to light of a selected wavelength;
- iv. tubing defining a sterile flow path between said separation chamber and said treatment container said flow path having length sufficient to allow for placement of said separation chamber within said separator and allow for placement of said treatment container is said separate irradiation device;
- v. a container of photoactive agent integral with said circuit, said container having an outlet in openable flow communication with an outlet flow path;
- and
- vi. a frangible cannula for preventing flow of photoactive agent from said container of photoactive agent to said treatment container,
- e) an automated cannula breaker configured to receive said cannula.
A method for combining a treating agent with a collected blood component comprising:
- a) mounting an integral disposable fluid circuit comprising a one or more vascular access devices, a separation chamber, a treatment container and a container of a treating agent on a separator unit and an irradiation unit wherein said separation chamber is mounted within said separator unit and said treatment container is mounted within said irradiation unit;
- b) introducing whole blood into said separation chamber;
- c) separating whole blood into blood components wherein one of said blood components comprises mononuclear cells;
- d) removing said mononuclear cells from said separation chamber;
- e) conveying said mononuclear cells from said separation chamber to said treatment container;
- and
- f) opening a flow path between said treating agent container and said treatment container.
