Abstract
Devices and methods for characterization of analyte mixtures are provided. Some methods described herein include performing enrichment steps on a device before expelling enriched analyte fractions from the device for subsequent analysis. Also included are devices for performing these enrichment steps.
Claims21
A method, comprising:
- introducing an analyte mixture into a microfluidic device containing a separation channel;
- applying an electric field across the separation channel to effect a separation of the analyte mixture;
- illuminating a first side of the microfluidic device through an optical slit;
- detecting light passing through the optical slit and the separation channel to monitor separation of the analyte mixture;
- and
- expelling a fraction of the analyte mixture from an orifice in fluid communication with the separation channel.
3 more elements
A method, comprising:
- introducing an analyte mixture into a microfluidic device including a separation channel;
- enriching a fraction of the analyte mixture in the separation channel;
- monitoring the separation channel while the fraction of the analyte mixture is enriched
- expelling the fraction of the analyte mixture from microfluidic device through an orifice disposed within a recess defined by the microfluidic device via electrospray ionization.
1 more element
A method, comprising:
- introducing an analyte mixture into a microfluidic device containing a separation channel loaded with an enrichment material configured to selectively bind to an analyte of interest contained within the analyte mixture;
- washing unbound analytes from the separation channel after the analyte mixture is introduced into the microfluidic device;
- introducing an eluent into the separation channel after the unbound analytes are washed from the separation channel such that the analyte of interest is eluted towards an orifice of the microfluidic device;
- and
- expelling the analyte of interest through the orifice via electrospray ionization, the orifice disposed within a recess defined by the microfluidic device such that a Taylor cone formed by the electrospray ionization is disposed entirely within the recess.
