Abstract
Compositions and methods are provided for genome modification at a target site in the genome of a filamentous fungal cell. The methods and compositions are drawn to a guide polynucleotide/Cas endonuclease system for modifying or altering the target site. Aspects in which the filamentous fungal cell being modified has a defective non-homologous end joining pathway are also provided.
Claims34
A method for homologous recombination of a donor DNA with a genomic locus in a filamentous fungal cell, the method comprising:
- a) introducing into a population of filamentous fungal cells a Cas endonuclease, a guide RNA, and a donor DNA comprising a domain with homology to a genomic locus of the fungal cell, wherein the Cas endonuclease and guide RNA are capable of forming a complex that enables the Cas endonuclease to act at a target site in or near the genomic locus of the fungal cells;
- and,
- b) identifying at least one fungal cell from the population in which homologous recombination of the donor DNA with the genomic locus has occurred, wherein the Cas endonuclease, the guide RNA, or both are introduced transiently into the population of fungal cells.
A recombinant filamentous fungal cell comprising a first recombinant DNA construct comprising an expression cassette for a Cas endonuclease.
