Abstract
Compositions, methods, and kits are disclosed directed at haptens, immunogens and immunoassays for oxycodone and metabolites thereof. The compounds are exemplified by compounds of the Formula I. The method comprises providing in combination in a medium (i) a sample suspected of containing oxycodone and/or oxycodone metabolites, a compound of the Formula I wherein R4 or R5 is a label, and an antibody for oxycodone or a metabolite thereof. The medium is examined for the presence of a complex comprising the labeled compound of Formula I where the presence of such as complex indicates the presence of oxycodone or oxycodone metabolite in the sample.

A compound of the Formula I:
- wherein:
- R1 is H or lower alkyl,
- R2 is H or OH, and
- R3 is —C(O)—(CH2)a—(NH—C(O)—(CH2)b)cR4, wherein a is an integer from 1 to 10, b is an integer from 1 to 10 and c is 0 or an integer from 1 to 5, and wherein R4 is halogen, an immunogenic carrier, or a label;
- or
- wherein R5 is an immunogenic carrier or a label and R6 is H, or wherein R5 and R6 are taken together to form a carbon-carbon double bond.
A method for determining an amount of oxycodone or a metabolite thereof in a sample suspected of containing oxycodone or a metabolite thereof, the method comprising:
- providing in combination in the medium:
- the sample suspected of containing oxycodone or a metabolite thereof;
- a compound of the Formula I:
- wherein:
- R1 is H or lower alkyl,
- R2 is H or OH, and
- R3 is —C(O)—(CH2)a—(NH—C(O)—(CH2)b)cR4, wherein a is an integer from 1 to 10, b is an integer from 1 to 10 and c is an integer from 1 to 5, and wherein R4 is a label;
- or
- wherein R5 is a label and R6 is H, and
- an antibody for oxycodone or a metabolite thereof;
- and
- examining the medium for an amount of a complex comprising the antibody and the compound of Formula I, the amount of the complex indicating the amount of oxycodone or a metabolite thereof in the sample.
A kit for determining an amount of oxycodone or a metabolite thereof in a sample suspected of containing oxycodone or a metabolite thereof, the kit comprising:
- a compound of the Formula I:
- wherein:
- R1 is H or lower alkyl,
- R2 is H or OH, and
- R3 is —C(O)—(CH2)a—(NH—C(O)—(CH2)b)cR4, wherein a is an integer from 1 to 10, b is an integer from 1 to 10 and c is an integer from 1 to 5, and wherein R4 is a label;
- or
- wherein R5 is a label and R6 is H;
- and
- an antibody for oxycodone or a metabolite thereof.
