Abstract
An optical measurement method and an optical measurement device for determining the spatial or spatiotemporal distribution of a sample, the sample comprising at least one retransmission source retransmitting light depending on light projected onto the sample according to a predetermined law. The method has steps of projection onto the sample of at least two compact light distributions belonging to different topological families, which propagate along the same optical path; detection of the light retransmitted by said at least one retransmission source of the sample; generation of at least one optical image from the detected light; and algorithmic analysis of the optical images for obtaining location data on said at least one retransmission source.
Claims13
A method of optical measurement for determining the spatial position of at least one luminous object in a sample, the method comprising:
- a. projecting onto the sample a sequence of a first compact luminous distribution and a second luminous distribution, wherein the first and second compact luminous distributions are of different topological families;
- b. projecting a sequence of compact light distributions relative to a hypothesized position of the at least one luminous object, each compact light distribution characterized by a null, in such a manner that there is a local minimum of luminosity at the null of each compact light distribution;
- c. detecting light re-emitted by the at least one luminous object in the sample;
- and
- d. obtaining spatiotemporal information with respect to the light re-emitted by the at least one luminous object on the basis of at least one of directly detecting a plurality of images and algorithmically analyzing the plurality of images.
A method of optical measurement for determining spatiotemporal information with respect to a set of luminous objects in a sample, the method comprising:
- a. projecting onto the sample a compact luminous distribution comprising a dark spot;
- b. dynamically determining a sequence of compact light distributions on the basis of at least one of a positioning hypothesis and a first set of measures;
- c. generating the sequence of compact light distributions;
- d. detecting light re-emitted by a blinking subset of the set of luminous objects in the sample;
- and
- e. obtaining spatiotemporal information with respect to the light re-emitted by the at least one luminous object on the basis of at least one of directly detecting at least one image and algorithmically analyzing the at least one image.
