Abstract
Disclosed are compositions of matters, cells, and treatment protocols useful for induction of anticancer responses in a patient suffering from cancer. In one embodiment the invention provides the use of NR2F6 silencing or gene editing in cord blood cells possessing anti-tumor activity in order to induce potentiated killer cells suitable for therapeutic use. In one embodiment said allogeneic cord blood killer cells are administered to initiate a cascade of antitumor immune responses, with initially responses mediated by allogeneic killer cells, and followed by endogenous immune responses.
Claims20
A method of inducing an anticancer immune responses comprising the steps of:
- a) obtaining a cord blood sample;
- b) isolated mononuclear cells from said cord blood sample;
- c) culturing said mononuclear cells in a means suitable for gene silencing of the NR2F6 immunological checkpoint;
- d) continuing said culture in the presence of factors capable of endowing and/or augmenting said cord blood mononuclear cells with tumor cytotoxic activity;
- and e) administering said cells to a patient in need of treatment.
The method of claim 84, wherein said enhanced immunogenicity is augmentation of HLA antigens.
